Go to the British Museum.
It is one of the best museums in the world. No jokes. They have the rosetta stone and tons of other stuff.
They also may have a major exhibition up in the central oval library thing. They have had in the past: Moctezuma (aztec/inca stuff), Terracotta army, Da Vinci and the Old masters (tons of sketchs that have never been shown outside private exhibits before), and lots of other stuff like that.
So you could get lucky and have one of those exhibits there while you are visiting.
The London Eye is really cool. Great view.
Half an hour in a pod looking at London. And if you are really feeling like treating yourself and whoever you may go with you could rent the pod for supper/drinks or something (but I think that is expensive and you would have to check and book).
Big Ben and the Houses of Parliament are interesting.
So is Buckingham Palace. Nice architecture around there.
Tower of London is also cool. Been there a few times on field trips and just visits.
The Royal Academy of Art is also really good.
They may have a good exhibit up. In the summer they always have a good exhibit. The Summer Exhibition. Artists will submit work each year for it, and it is always good and interesting.
Across the road from the RAA is
Fortnum and Mason.
I worked there for 2 and a half months and it is really fancy.
Best place for tea in england. Also great biscuits and chocolate. Great to get presents there as they are quintescentially english.
Also out of Harrods and F&M, F&M is the only one to have the Queen's Charter (Harrods had it but lost it).
The Ritz is right down the road along with the Worsley (a really fancy resteraunt).
If you want great resteraunts from all styles of extremely fancy to just great food and a decent price I can tell you.
Also cheese shops, wine shops and stuff like that.
If you have any kids that you are related to a trip to Hamleys is a good choice. A few toys from there will make you favourite whatever (parent, uncle, etc.)
Tons of funky clothes shops in Notting Hill. So maybe some steampunk shops, retro, indie, etc. the list goes on.
Whatelse have I left out?
There is a lot of london that people forget about that isn't
that touristy.
And even the touristy places can be not that touristy if you are going outside of holiday times.
So like the Tower of London. If you are in the lull season, it is a must see.
You can easily fit in a majority of what I have said into a single day. If you plan ahead first and are pacy with your transition from one place to another. (I.e. I could easily hit a number of what I said in one day and still take it all in).

Oh before I forget. SOHO, Leicester Square (big movie theatre square), and Chinatown.
And also Theatres and Concert Halls. There are tons of those. So musicals, plays, operas, ballet, etc.
Anything you can really think of you will be able to find in London.
So as someone said earlier, give us stuff to work with and we will help out.

OH and one more thing.
I misread your first post and tomorrow I will endeavor as soon as I can to give you possibilities around london and far from london (depending on your choice) that you could go and see and visit.
I know quite a bit about a few of the other english cities and landmarks.
If you are going through the countryside by car go to Heston Blumenthals resteraunt. He is a crazy chef, but frickin' amazing. If you order seafood then you listen to the sound of the sea while you eat. There is bacon ice cream. Etc. List goes on. Google him.
He has almost as many chefs in his resteraunt as he gets customers. And he has a waiting list I think.
Wooopps. A bit more than one thing, but oh well, and extra minute helping out a friend isn't wasted in my book.
